چکیده

Identification of all genes involved in the phytochrome (phy)-mediated responses of plants to their light environment is an important goal in providing an overall understanding of light-regulated growth and development. This article highlights and integrates the central findings of two recent comprehensive studies in Arabidopsis that have identified the genome-wide set of phy-regulated genes that respond rapidly to red-light signals upon first exposure of dark-grown seedlings, and have tested the functional relevance to normal seedling photomorphogenesis of an initial subset of these genes. The data: (a) reveal considerable complexity in the channeling of the light signals through the different phy-family members (phyA to phyE) to responsive genes; (b) identify a diversity of transcription-factor-encoding genes as major early, if not primary, targets of phy signaling, and, therefore, as potentially important regulators in the transcriptional-network hierarchy; and (c) identify auxin-related genes as the dominant class among rapidly-regulated, hormone-related genes. However, reverse-genetic functional profiling of a selected subset of these genes reveals that only a limited fraction are necessary for optimal phy-induced seedling deetiolation.

Identification of a cis-regulatory element involved in phytochrome down-regulated expression of the pea small GTPase gene pra2.

The pra2 gene encodes a pea (Pisum sativum) small GTPase belonging to the YPT/rab family, and its expression is down-regulated by light, mediated by phytochrome. We have isolated and characterized a genomic clone of this gene and constructed a fusion DNA of its 5'-upstream region in front of the gene for firefly luciferase. Using this construct in a transient assay, we determined a pra2 cis-reg...

Phytochrome-regulated repression of gene expression requires calcium and cGMP.

The plant photoreceptor phytochrome A utilizes three signal transduction pathways, dependent upon calcium and/or cGMP, to activate genes in the light. In this report, we have studied the phytochrome A regulation of a gene that is down-regulated by light, asparagine synthetase (AS1). We show that AS1 is expressed in the dark and repressed in the light. Repression of AS1 in the light is likely co...

EARLY FLOWERING 4 functions in phytochrome B-regulated seedling de-etiolation.

To define the functions of genes previously identified by expression profiling as being rapidly light induced under phytochrome (phy) control, we are investigating the seedling de-etiolation phenotypes of mutants carrying T-DNA insertional disruptions at these loci. Mutants at one such locus displayed reduced responsiveness to continuous red, but not continuous far-red light, suggesting a role ...

Light-regulated nuclear localization of phytochromes.

Phytochrome is a soluble protein that regulates various responses of plants to light. Not all but most of the phytochrome responses are accompanied by changes in the pattern of gene expression. Upon light activation, phytochrome is imported into the nucleus by the nuclear localization activity of the carboxy-terminal half of the molecule. In darkness, the amino-terminal chromophoric domain supp...

Different Roles for Phytochrome in Etiolated and Green Plants Deduced from Characterization of Arabidopsis thaliana Mutants.

We have isolated a new complementation group of Arabidopsis thaliana long hypocotyl mutant (hy6) and have characterized a variety of light-regulated phenomena in hy6 and other previously isolated A. thaliana hy mutants. Among six complementation groups that define the HY phenotype in A. thaliana, three (hy1, hy2, and hy6) had significantly lowered levels of photoreversibly detectable phytochrom...

Different roles for calcium and calmodulin in phytochrome- and UV-regulated expression of chalcone synthase

Based on knowledge of white light-mediated gene induction events in a photomixotrophic soybean cell culture, a comparative pharmacological dissection of gene expression in response to irradiation with red and UV light was performed in order to examine the molecular basis of cross-talk between phytochromeand UV-regulated signalling pathways. This has proved particularly interesting for the gene ...
